一種高酸鹼反應範圍之聚木醣之突變基因及其點突變方法,其係利用該點突變方法將一聚木醣之基因之第41個氨基酸由天門冬醯胺[asparagine]突變為天門冬胺酸[aspartic acid],以形成該高酸鹼反應範圍之聚木醣之突變基因。藉此,可有效提高該聚木醣之酸鹼耐受性、酸鹼反應範圍及活性。本發明之目的係高酸鹼反應範圍之聚木醣酶之突變基因的第41個氨基酸係為天門冬胺酸,使得本發明具有在不降低反應活性下提高一聚木醣酶之酸鹼反應範圍的功效,並將一酵素之基因中的至少一氨基酸由天門冬醯胺突變為天門冬胺酸,以形成該酵素之一突變基因,進而使得本發明具有提高該酵素之酸鹼反應範圍及活性之功效。
